Stephen and Christine dated prior to 2016 but broke up due to his arogant behaviour, but they still managed to work together in the hospital. After Stephen got into a car accident, Christine was the one who looked after him but left after he became rude to her and said goodbye and after he had to quit being a doctor due to a fracture in his hands. The two saw each other again one year later when Stephen visited Christine at the hospital where he asked for her help with his injuries. Strange told Palmer that he had joined the Mystic Arts which caught him magic and she operated on him, saving his life as he was dying from his injuries. After Stephen was able to prove to Christine that he was telling the truth on learning magic, they rekindled their friendship but didn't get back together.
While Stephen was a victim of Thanos' snap in 2018, Christine survived and met another guy whom she married, named Charlie, inviting Stephen to their wedding. After the ceremony, Stephen apologises to Christine for not being there for her all those years ago but is glad she is happy. When he and America Chavez find themselves in a parallel world, Stephen meets an alternate Christine who reveals it did not work out between her and her Doctor Strange who had died. Although reluctant to help another version of her late boyfriend, Christine agrees and becomes close with this Stephen as a result. It is also revealed that there are multiple versions of Doctor Strange throughout the multiverse and Christine's, but the relationships don't work out. After Scarlet Witch's defeat, Stephen finally gets closure with Christine and tells this alternate version that he loves her in every universe. Touched, Christine tells Stephen to face his fears about relationships and it allows Stephen to finally move on.

Wong
Wong is Stephen's best friend who trained him when he first came to Karma Taj and helped him fight the villain. Wong let Stephen reside in the New York Sanctum after realising he is capable of using magic. The two became good friends over the years and often worked alongside each other during attacks. Wong also took over Stephen's role as Sourcer Supreme after he became a victim of Thanos' snap in 2018. Stephen eventually warmed up to the idea of his friend becoming his boss.
Tony Stark/Iron Man
Doctor Strange met Tony in Avengers: Infinity War, when the magician transported himself to New York to discuss the upcoming attacks with Stark. Upon meeting the sorcerer, Stark imminently made fun of Strange and that he looked like somebody about to perform magic tricks at a child's birthday party. Ignoring Stark's insults, Strange informed Stark that he and Bruce Banner needed his help, so portaled the hero to the sanctum, where Wong caught Stark the importance of the Infinity Stones and what could happen if Thanos got all 6 stones.
Despite their occasional bickering, the duo tried to put their differences aside when they realised two of Thanos' henchmen had arrived in Grenich Village for the time stone. Strange and Stark worked together with Wong, and later Spider-Man, who had blown off his field trip to help the Avengers. Stark tried to save Strange, but Ebony Maw managed to capture Strange temporally and they ended up on the Q-ship with Spider-Man, where Stark and Parker managed to save Strange's life by sending Ebony Maw into the galaxy and freeze to death. However, Strange was less than thankful for Stark saving him, but they managed to pilot the ship to Thanos' planet, Titan, with Spider-Man to confront Thanos directly, where they met the Guardians of the Galaxy, a team whom became their allies after realising both teams were after Thanos. Stark did show concerns for Strange when he noticed the sorcerer zoned out and seeing 14, 000, 605 possible outcomes to their current battle. At the end of the battle of Titan, Doctor Strange willingly gave Thanos the time stone to spare Stark's life. After Thanos teleported away with the time stone, a wounded Iron Man asked Strange why he gave the mad titan the stone, to which Strange said that there was no other way in the possible outcomes he saw. With that, Strange became one of the victims obliviated by Thanos' snap, while Stark survived.
Five years later in 2023, Stark and the remaining, original 6 avengers, managed to undo Thanos' snap, which resurrected all the victims who were killed five years later, which included Strange. During the battle, Stark asked him if this battle was part of one of the outcomes Strange saw, to which the sorcerer mentioned he could not say anything because it could have lead to the heroes losing again. However, later in the brutal battle, Strange signalled Iron Man to let him know that this indeed was the plan and the one outcome the heroes would win, but it would mean Stark would have to use the stones himself. Stark accepted his fate and obliterated Thanos and his army from existence with his own snap, with the cost of his own life. Strange joined the resurrected heroes in kneeling to the fallen hero as Iron Man died on the battlefield, and attended his funeral.
Peter Parker/Spider-Man

Peter met Stephen Strange met during Thanos' attack in Avengers: Infinity War after the former and Tony Stark saved his life from Ebony Maw, who was about to kill him to take his Time Stone. At first, Strange and Peter were nothing more than situational allies and Strange was even initially willing to let Parker or Stark die in the battle against Thanos to protect the Time Stone from him. Strange's decision to spare Tony in exchange for Thanos getting the Time Stone ultimately resulted in his and Peter's deaths after Thanos wiped out 50% of the population. However their relationship became better following Avengers: Endgame, with Parker seeking Strange's help during the events of Spider-Man: No Way Home to cast a spell to make the world forget he is Spider-Man to take off heat from his friends, Aunt May and Happy Hogan. Their friendship has improved to the point where he tells Peter that he does not need to address him formally. Peter ruins the spell however, and opens the multiverse, annoying and angering Strange to a great degree with the latter telling him bluntly that his double life was the cause of all his problems. Peter and Strange eventually clash after the latter callously decides to send the multiversal villains home to their demises at the hands of their Spider-Man, which the former disagrees with and stops him from doing so. Peter then traps Strange in the Mirror Dimension. Later, after being released and following Peter having cured the villains, Strange is asked by Peter to cast a spell to forget Peter Parker to save their universe from collapsing, which he hesitates at first, as he had come to see Peter as a friend and did not want to forget him in addition to not wanting to ruining his life further, but ultimately accepts and respects Peter's decision, even telling him he can call him by his first name which he thought was weird at first. Strange is secretly grateful that Peter helped him find his own lost goodness and subconsciously held on to his teachings as he tried to protect and foster a friendship with America Chavez and give Wanda Maximoff multiple chances to redeem herself.

Doctor Strange first meets America Chavez when he notices she is being chased by a monster across New York city while attending Christine Palmer's wedding. Stephen manages to save America but she fears him at first because of his alternate self betraying her. They discuss her powers, but she is scared to trust him due to his alternate self. Stephen constantly tries to protect America, which results in them landing in a universe known as Earth-838 and they discuss his relationship with Christine, where America tells Stephen the version of him she knew blew it with his version. Learning that the Scarlet Witch is after America for her powers, Doctor Strange protects her against the witch. The duo quickly become close and when Strange and an alternate Christine find themselves in a damaged universe, he uses the dreamwalking power to possess his alternate self to convince Chavez that she can use her power against Wanda. After Scarlet Witch's death, America manages to find Strange and Palmer in Sinster Strange's reality and brings them back to their respective universes. Strange and Wong allow Chavez to stay in Karma-Taj for a while where she can be safe and train while Strange starts to return to the Sancum. Before he leaves, America tells Stephen that she is glad she fell into his universe, which he reciprocates.
Wanda Maximoff/Scarlet Witch
In Doctor Strange in the Multiverse of Madness, Doctor Strange visited Wanda at her home to ask her for help regarding the safety of America Chavez. Wanda thought Stephen wanted to talk about her recent actions in Westview but he looked past that, seeing the good in her and that she made mistakes with the town. However, Strange soon learnt that Wanda was the reason that America was being hunted by the monsters after planning to use her to travel to another universe to be with her sons, and was planning on stealing her powers to do so. Strange learnt that the Darkhold had corrupted Wanda's mind and that she had become the villain known as the Scarlet Witch. Strange and the Masters of the Mystic Arts tried to reason with the witch but she refused to listen, calling him a hypocrite for giving Thanos the time stone and that Vision died. Seeing what Wanda had become, Strange tried to protect her alternate self and America from her. Dream walking into his variant's corpse, Strange and Chavez managed to make Wanda realise what she had become which caused her to destroy the Darkhold in every universe and saw that there was truly goodness in her after all.

Doctor Strange fought Kaecilius when he first started out as a sourcerer and managed to defeat him when he was turned into a mindless being by the Ancient One.
Thanos
Doctor Strange learnt of Thanos from Bruce Banner after the hero crash landed into the New York Sanctum in 2018. Strange learnt that Thanos wanted the Time Stone to complete his gautlet to erase half of the universe. Strange banded with Iron Man and Spider-Man to stop Thanos from completing his mission, along with the Guardians of the Galaxy. Strange saw 14, 00, 605 possible futures where the Avengers could win but only saw one. Seeing Iron Man in danger, Doctor Strange willingly gave Thanos the time stone to spare Stark's life, which partially meant Thanos could succeed in destroying half of the universe. As he died, Strange told Thanos Iron Man that giving Thanos the time stone was the only way in the possible futures. Five years later, Strange took part in the Battle of Earth against Thanos and watched as Iron Man managed to erase the Mad Titan from existance, at the cost of Stark's life.
