Direct chemical evidence for sphingolipid domains in the plasma membranes of fibroblasts

Abstract: Sphingolipids play important roles in plasma membrane structure and cell signaling. However, their lateral distribution in the plasma membrane is poorly understood. Here we quantitatively analyzed the sphingolipid organization on the entire dorsal surface of intact cells by mapping the distribution of 15 N-enriched ions from metabolically labeled 15 N-sphingolipids in the plasma membrane, using highresolution imaging mass spectrometry. Many types of control experiments (internal, positive, negative, and fixati… Show more

“…In retrospect, this probe of endogenous SM validates our previous observations on SM submicrometric domains using insertion of exogenous BODIPY-SM into partially spread nonfi xed RBCs. This main conclusion, derived from two vital imaging tools, is also entirely consistent with recent evidence by highresolution methods in fi xed cells ( 8,31,38,41 ). Second, we provide basic mechanistic insights on domain composition and regulation, based on lipid phase transition (temperature), cholesterol, and membrane tension.…”

“…By electron microscopy, SM-rich domains labeled with lysenin on fi xed Jurkat cells were ‫ف‬ 150 nm in diameter ( 38 ). High-resolution ion mass spectrometry based on 15 NSLs revealed domains of ‫ف‬ 200 nm in diameter in fi xed fi broblasts, interestingly regulated by cholesterol and actin cytoskeleton ( 8 ). While it is unclear whether these different values result from alternative methods or cells, all previous researches, combined with this study, have concurred to establish the concept of rather stable submicrometric SM domains on eukaryotic cells, thus conceptually distinct from transient nanometric rafts.…”

“…Addressing this will necessitate better assays to relate the local composition of the membrane milieu to the clustering of GPI-anchored proteins. Multimodal approaches combining the measure of local lipid order ( 127-129 ) and clustering or imaging-mass spectroscopy tools like NanoSIMS ( 130,131 ) could be key.…”

“…This imaging modality achieves chemical specificity by using isotopic labeling of _target molecules and affords high spatial resolution up to 50 nm. Chemoselective and quantitative imaging modalities based on MS are also expected to bring a new dimension to lipid raft research (41).…”
