Autophagy Pathway Is Required for IL-6 Induced Neuroendocrine Differentiation and Chemoresistance of Prostate Cancer LNCaP Cells

Abstract: Prostate cancer (PCa) cells undergoing neuroendocrine differentiation (NED) are clinically relevant to the development of relapsed castration-resistant PCa. Increasing evidences show that autophagy involves in the development of neuroendocrine (NE) tumors, including PCa. To clarify the effect of autophagy on NED, androgen-sensitive PCa LNCaP cells were examined. Treatment of LNCaP cells with IL-6 resulted in an induction of autophagy. In the absence of androgen, IL-6 caused an even stronger activation of autop… Show more

“…In the same view, Chang et al [61] showed that autophagy is involved in PCa progression. In addition, autophagy seems to play a cytoprotective role in IL-6 induced NE differentiation, and inhibition of autophagy by knockdown of beclin1 or Atg5 sensitizes NE differentiated LNCaP cells to chemotherapy with etoposide.…”

“…CXCR1 is overexpressed in PCa cells, and it has been hypothesized that paracrine activation of CXCR1 by IL-8 may contribute to androgen-independent proliferation of prostate cancer in patients treated with ADT [61]. Moreover, NE cells express IL-8 and CXCR2, as observed by staining serial sections of PCa tissue [64].…”

“…Interleukin 6 has also been studied, and can function to activate both MAPK and PI3K pathways, and its implications have been seen in PCa (Chang et al, 2014). Up-regulation of this extracellular protein was seen in both prostate tissue and patient serum of hormone refractory disease (Chang et al, 2014).…”

“…25 Another study implemented on neurendocrine differentiation in prostate cancer cells revealed that knockdown of REST activates the IL-6-induced autopaghy pathway. 26 The authors also showed that IL-6-induced neuronal cell morphology changes in prostate cancer cells were significantly inhibited when there was REST overexpression. This finding was confirmed in a subsequent study by showing that the overexpression of exogenous REST abrogates IL-6-induced neuroendocrine differentiation in prostate cancer cells.…”
