MG53-mediated cell membrane repair protects against acute kidney injury

Abstract: Injury to the renal proximal tubular epithelium (PTE) represents the underlying consequence of acute kidney injury (AKI) after exposure to various stressors, including nephrotoxins and ischemia/reperfusion (I/R). Although the kidney has the ability to repair itself after mild injury, insufficient repair of PTE cells may trigger inflammatory and fibrotic responses, leading to chronic renal failure. We report that MG53, a member of the TRIM family of proteins, participates in repair of injured PTE cells and prot… Show more

“…Hydrogel Encapsulation Provides Efficient Delivery of rhMG53 to Dermal Wounds-Pharmacokinetic studies have shown that rhMG53 has a short half-life of ϳ1.5 h in the blood circulation after intravenous or subcutaneous administration (18,21). Therefore, treatment of open dermal wounds required repetitive subcutaneous administration of rhMG53.…”

“…Moreover, the recombinant human MG53 (rhMG53) protein can protect various cell types against membrane disruption when applied to the extracellular environment and ameliorate the pathology associated with muscular dystrophy and lung injury (18,19). We have also shown that rhMG53 is effective in protection against myocardial infarction (20) and acute kidney injury (21).…”

“…It has been shown that in skeletal muscle cells breakage of the plasma membrane causes overexpression of Trim72 at the injury site, accompanied by a reduced extracellular dye penetration into the cell, suggesting the formation of a repair patch by Trim72 [28,29] . The tripartite motif proteins are large and display diverse cell biological functions.…”

“…MG53 consists of an amino-terminal TRIM domain and a carboxy-terminal SPRY domain, a common structure of most TRIM family proteins. RNA analysis revealed MG53 is highly expressed in striated muscle and to a lower extent in lung and kidney epithelia (Cai et al, 2009a; Duann et al, 2015; Jia et al, 2014). Because of its high expression in skeletal muscle, MG53 was investigated as a potential mediator of membrane repair at the site of injury (Cai et al, 2009a).…”
